Processing Optimization Tips
- Controlling Contrast: If you prefer darker, more pronounced nuclear detail, you can easily increase the dipping count in Solution II (e.g., 7–8 dips). If the background or cells appear overly blue, decrease the Solution II dipping time or increase your final water rinse.
- Water Contamination: Keep the lid on the Fixative container tightly sealed when not in use. Methanol absorbs ambient humidity rapidly, and excessive water contamination will cause cell distortion and poor fixation artifacting over time.
