A complete Gram Colour Kit features four primary diagnostic reagents, typically arranged in sequence: [1]

- Mordant (Gram’s Iodine Solution): A chemical fixative that forms a large, water-insoluble complex with the crystal violet inside the cell. [1, 2, 3, 4, 5]
- Decolourizer Solution (Acetone-Alcohol): A solvent mixture used to selectively strip the primary dye-mordant complex out of specific bacterial walls. [1, 2]

Staining Mechanism & Principle
- Gram-Positive Bacteria: These organisms possess a thick, highly cross-linked peptidoglycan cell wall. When the iodine mordant is applied, it creates a massive “dye lake” complex inside the cell. The acetone-alcohol decolourizer dehydrates this thick cell wall, causing it to shrink and trap the large dye-iodine complex inside. As a result, Gram-positive cells retain the primary stain and appear deep blue to purple under the microscope. [1, 2, 3, 4, 5]
- Gram-Negative Bacteria: These organisms have a much thinner peptidoglycan layer surrounded by an outer lipopolysaccharide membrane. The alcohol-acetone decolourizer easily dissolves the lipid-rich outer membrane and creates large pores in the thin peptidoglycan layer, allowing the blue dye-iodine complex to wash out entirely. Once colorless, these cells absorb the Safranin counterstain, causing them to appear pink to red. [1, 2, 3, 4, 5]
Microscopic Visual Summary
When evaluated under a high-power oil immersion lens (100x), the visual presentation yields distinct diagnostic clues:
- Gram-Positive Cells: Dark purple/blue (e.g., Staphylococcus aureus, Streptococcus pneumoniae).
- Gram-Negative Cells: Pink/red (e.g., Escherichia coli, Pseudomonas aeruginosa).
