Description
The India Ink Solution provided by IBEX SCIENCES is a high-purity, stable colloidal suspension of carbon black particles. This reagent is utilized for two entirely distinct purposes depending on whether it is deployed in a microbiology lab or a surgical histopathology suite. [1, 2]

1. Microbiological Application: Negative Staining
In diagnostic microbiology, India Ink is the gold standard for performing a negative stain to visualize encapsulated microorganisms.
  • The Principle: The carbon particles in India Ink are relatively large and possess a negative surface charge. Because bacterial and fungal cell walls (and capsules) also carry a negative charge, they repel the ink.
  • The Visual Result: The ink cannot penetrate the capsule. Under a microscope, the background appears dark and opaque, while the encapsulated organisms stand out as clear, glowing, halo-like structures. [1]
  • Primary Target: It is clinically vital for diagnosing cryptococcal meningitis by identifying Cryptococcus neoformans in cerebrospinal fluid (CSF) sediments. [1, 2, 3]
2. Histopathological Application: Surgical Margin Inking
In anatomical pathology, India Ink is applied directly to the outer surface of fresh or fixed surgical tissue specimens (such as tumor biopsies or lumpectomies).
  • The Principle: The carbon particles bind strongly to the tissue surface, leaving a permanent dark border that remains completely intact through routine tissue processing, paraffin embedding, and slide sectioning.
  • The Visual Result: Under the microscope, the pathologist can clearly see where the true surgical edge resides. This allows them to accurately measure the distance between a malignant tumor and the resection margin to confirm if the cancer was entirely removed.

Standard Diagnostic Method (For CSF Testing)
  1. Sample Preparation: Place a drop of centrifuged CSF sediment or a loopful of culture on a clean glass slide.
  2. Reagent Application: Add an equal drop of India Ink Solution directly next to the sample.
  3. Mixing: Gently mix the drops together using a loop or the edge of a coverslip, then apply a coverslip.
  4. Microscopic Review: Examine the slide under a standard light microscope using low and high-power objectives to look for distinct, clear halos surrounding budding yeast cells.
Best Practices for Laboratory Use
  • Check for Contamination: Over time, open bottles of India Ink can harbor saprophytic fungi or bacteria. Periodically check the solution under a microscope to confirm the background is free of artifacts.
  • Storage Requirements: Keep the solution container tightly sealed at standard room temperature (15°C to 25°C). Avoid freezing, as freezing can cause the carbon particles to clump together and precipitate out of the suspension.
