Kit Components
- Fixative Solution: A methanol-based reagent that fixes the air-dried smear to the slide.
- Solution I (Eosinophilic/Acidic Stain): A red buffered solution that stains alkaline components like the cytoplasm and red blood cells.
- Solution II (Basophilic/Alkaline Stain): A blue azur/methylene blue solution that stains acidic components like nuclear chromatin.

Typical Procedure
- Step 1: Prepare a thin blood smear and allow it to completely air-dry.
- Step 2: Dip the slide into the Fixative Solution for 5–10 seconds.
- Step 3: Immerse the slide into Solution I for approximately 5–10 seconds.
- Step 4: Immerse the slide into Solution II for approximately 5–10 seconds.
- Step 5: Briefly rinse the slide in the buffer solution or deionized water, drain vertically, air-dry, and examine under a microscope. [1, 2, 3, 4, 5]
