Core Principle and Mechanism
When bacteria are cultivated in a medium rich in tryptophan (such as Peptone Water or Tryptone Broth), those that produce tryptophanase split the tryptophan molecule to yield indole, pyruvic acid, and ammonia. [1, 2, 3]
When IBEX Indole Test Reagent is added to the broth, the isoamyl alcohol extracts the indole to the surface. There, the indole reacts with the DMABA under acidic conditions, forming a distinct quinoid dye.
- Positive Result: Formation of a distinct, deep cherry-red ring in the alcohol layer at the top of the broth within a few seconds. This indicates the organism produces indole (e.g., Escherichia coli, Proteus vulgaris). [1, 2, 3]
- Negative Result: The top layer remains the original yellow or amber color of the reagent, indicating no indole was produced (e.g., Klebsiella pneumoniae, Salmonella species).
Step-by-Step Test Procedure
- Inoculation: Inoculate a tube of Tryptone or Peptone broth with a pure colony of the test bacterium.
- Incubation: Incubate the broth at 35°C–37°C for 24 to 48 hours to allow sufficient bacterial growth and tryptophan metabolism.
- Reagent Addition: Directly add 0.5 mL (approx. 5 to 10 drops) of the IBEX Indole Test Reagent to the culture tube.
- Agitation & Observation: Shake the tube gently to facilitate extraction, then let it stand upright. Observe the immediate color change in the rising surface fluid layer. [1, 2]
Crucial Practical Limitations
- Media Requirements: The test must be performed from a broth medium specifically containing tryptophan. Media like Glucose broth or general nutrient broth without optimal peptone levels will yield false negatives. [1]
- Light Sensitivity & Stability: Kovac’s reagent is highly sensitive to light and temperature. Over time, it can undergo auto-oxidation and turn a dark brown color. If the reagent in the bottle loses its pale yellow/clear hue and becomes dark, it has deteriorated and must be discarded.
- Corrosive & Fuming Hazard: Due to the concentrated hydrochloric acid and isoamyl alcohol content, the reagent is highly corrosive, toxic, and flammable.
