The IBEX Sciences PAS Colour Kit (Periodic Acid–Schiff Staining Kit) is a high-performance histochemical staining system designed to demonstrate carbohydrates, polysaccharides (such as glycogen), and mucosubstances (like glycoproteins, glycolipids, and mucins) in tissue sections. [1]
Manufactured by IBEX Sciences, this specialized kit is a vital analytical tool in histopathology, hematology, and medical research for diagnosing glycogen storage diseases, detecting mucin-producing tumors, evaluating renal glomerular basement membranes, and identifying fungal pathogens. [1, 2]

Core Kit Components
The multi-reagent system typically includes three primary components designed to work in a carefully timed sequence: [1]
  1. PAS Oxidant Reagent (Periodic Acid Solution): An aqueous solution used to chemically initiate the reaction by breaking specific carbon-carbon bonds within standard carbohydrate rings. [1, 2]
  2. Schiff Dye Solution (Feulgen Reagent): A specialized formulation containing basic fuchsin and sulfurous acid that targets newly exposed cellular components to deposit a highly intense, stable pigment. [1, 2]
  3. Counterstain (Typically Harris or Mayer’s Hematoxylin): A basic dye that targets structural nucleic material to give back-end tissue contrast. [1, 2]

Chemical Operating Principle
The histochemical mechanism occurs in two distinct, sequential phases:
  • Phase 1 (Oxidation): The periodic acid solution oxidizes the adjacent glycol groups (vicinal diols) of the carbohydrates, converting them into 1,2-aldehydes. This process must be carefully controlled to ensure the aldehydes are not over-oxidized. [1, 2]
  • Phase 2 (Color Development): The tissue section is introduced to the Schiff reagent. The free aldehyde groups react directly with the colorless fuchsin-sulfurous acid compound. This re-establishes the chemical chromophore, resulting in an irreversible condensation product that stains the carbohydrate-dense areas a vibrant magenta. [1, 2, 3]

Microscopic Visual Interpretation
When properly processed and viewed under a laboratory microscope, cellular elements exhibit high contrast differentiation: [1]

PAS-Positive Materials (Glycogen, Mucins, Basal Membranes) Bright Magenta / Purple-Red Identifies cellular glycogen deposits, secretory neutral mucins, and intact or thickened basement membranes.
Fungal Organisms (Candida, Aspergillus) Brilliant Magenta The chitin-and-cellulose-dense cell walls of fungal hyphae or budding yeasts stain intensely, allowing for easy clinical screening.
Cell Nuclei Blue / Black The hematoxylin provides an optimal background contrast to evaluate overall cellularity and nuclear features.


Standard Lab Workflow Guidelines
  1. Deparaffinization: Deparaffinize and hydrate formalin-fixed, paraffin-embedded (FFPE) tissue sections down to deionized water. [1, 2]
  2. Oxidation: Submerge the slides in the Periodic Acid Solution for roughly 5 to 10 minutes at room temperature, then rinse thoroughly with running water to remove excess oxidant. [1, 2]
  3. Schiff Reaction: Immerse the slides into the Schiff Reagent for 15 to 30 minutes. (A distinct, deep magenta color tone will develop across the tissue section). Rinse thoroughly under running water. [1, 2]
  4. Counterstaining: Counterstain the nuclei using the Hematoxylin Solution for 2 to 5 minutes, follow with a brief bluing step, wash, dehydrate through graded alcohols, clear in xylene, and mount for microscopy. [1, 2, 3, 4]

Handling and Storage Best Practices
  • Temperature Restrictions: The Schiff reagent component is chemically volatile and light-sensitive. It must be stored under refrigeration between 2°C and 8°C to maintain structural stability, while the remaining components can be kept at ambient room temperature.
  • Deterioration Indicators: The Schiff reagent should remain clear and colorless. If the solution inside the bottle develops a pink or amber discoloration before being exposed to tissue aldehydes, it has auto-oxidized and must be discarded.
  • Carry-Over Prevention: Always drain excess water off the slides between steps to prevent diluting the primary reagents, and never reuse aliquoted working reagents back into the master IBEX Sciences stock bottles. [1, 2, 3, 4]
